Avian retrotransposons enabled targeted DNA integration

Figure 1: Yangcan Chen et al. Discovery and engineering of avian R2 retrotransposons for all-RNA-mediated targeted DNA integration in human cells. Nature Biotechnology (2026).Full-size image

A Nature Biotechnology study identified 159 R2 retrotransposons by searching 1,139 avian genomes. The authors characterized conserved and nonconserved features in their proteins and untranslated regions. Engineered variants achieved up to 60% site-specific gene integration across human primary cells. The sequencing data were deposited in the Genome Sequence Archive for Human under accession HRA013312.

The resource may help genome-editing researchers develop all-RNA integration tools. The reported results were in human primary cells.
